Abstract
Background: This study was conducted to determine the association between polymorphism of interleukin (IL)-10 -1082G/A gene on susceptibility of multidrug resistant tuberculosis (MDR-TB). Polymorphism of IL-10-1082G/A as a single nucleotide polymorphism of IL-10 gene affects IL-10 cytokine production. The IL-10 cytokine is a potent immunoregulator to TB infection. Polymorphism of IL-10 -1082G/A gene is susceptible for TB.
Methods: Socio-epidemiological data and blood specimens were collected from 30 MDR-TB subjects, 29 non MDR-TB subjects and 34 healthy subjects. Blood specimens were analyzed for IL-10-1082G/A gene polymorphism by PCR. This study was a cross-sectional study conducted in Dr Moewardi Hospital Surakarta on November and December 2014. We compared the frequencies of IL-10-1082G/A, IL-10-1082A/A, and IL-10-1082G/G genotypes among groups.
Results: Genotype of IL-10 -1082 A/A was more frequent in MDR-TB group 33.3% (10/30) compares to non MDR-TB group 10.3% (3/29) and healthy group 8.8% (3/34). The frequency of IL-10-1082G/A genotype in MDR-TB group 66.7% (20/30), TB group 89.7% (26/29), and healthy group 91.2% (31/34). The frequency of IL-10-1082A/A and IL-10 -1082 G/A genotypes were significantly different between these groups (p=0.051). Polymorphism of IL-10-1082G/A gene is associated to MDR-TB. There was no IL-10-1082G/G find in this study.
Conclusion: Polymorphism of IL-10-1082G/A gene is associated to susceptibility of MDR-TB in this study. (J Respir Indo. 2016; 36: 1-10)
